Super-resolution optical microscopy elucidates cellular structure beyond the diffraction limit,

enabling discoveries in the life sciences. Cryogenic optical localization provides further depth,

exploiting the enhanced photo-stability of organic fluorophores at low temperatures and their

random on-off switching to determine the position of several labelling sites with Angstrom

precision. Therefore, the method advances optical microscopy to the three-dimensional molecular

architecture of proteins and bio-molecular assemblies.

In this talk, we start with the detection of single-molecules with an optical microscope, demonstrate

how we achieve super-resolution via precise localization, and go on to explore potential

applications. The polarization of the emitted fluorescence light is included as an additional

parameter in the identification and localization of several molecules within a diffraction-limited

area. An application for determining inter-molecular distances of a few nanometers is discussed

We also show that we can combine this approach with image classification to address

heterogeneous samples. Lastly, we present a workflow for the preparation and imaging of vitrified

specimens adapted to our optical microscope, allowing quantitative information on the structure

of macro-molecules in a near-native state and environment to be obtained. Exciting prospects are

correlative studies with cryogenic electron microscopy and tomography, particularly of membrane
